  1. In the paper Soldi and Bonaldi describe a novel analytical method to dissect the proteomic composition of chromatin, named ChroP (Chromatin Proteomics). ChroP combines a preparative version of Chromatin immunoprecipitation (ChIP) to enrich distinct chromatin regions, with Mass Spectrometry-based quantitative proteomics to analyze its composition, in terms of histone PTMs and variants, transcription factors and multi-protein complexes specifically co-associating. The paper illustrates how ChroP approach enables to characterize the synergisms between these determinants at functionally distinct chromatin domains, with a resolution of mono- to oligo-nucleosomes.
    The approach is relatively easy to be implemented in the epigenetics groups, given the limited changes made to conventional N- and X- ChIP protocols, used in ChIP-Seq studies. Hence, for this rather straightforward optimization, ChroP will be amenable to various applications in functional studies, such as the analysis of the dynamic changes of chromatin structure upon distinct perturbations (i.e. global transcriptional activation, differentiation, depletion of nuclear factors, treatment with epigenetic drugs, etc.). TB group is available for counseling to setup and optimize ChroP for application in such studies.
    ChroP represents an additional and valuable tool in the arsenal of analytical strategies available to study chromatin composition and dynamics, in a system-wide fashion.
